How does Barcoding Technology Work?
The multiplexing capability of the PhenoCycler-Fusion 2.0 system is underpinned by the fundamental principles of barcoded antibody technology. In this framework:
- Each PhenoCycler antibody is intricately linked to a unique oligonucleotide sequence, known as the PhenoCycler Barcode.
- Each PhenoCycler Reporter is a fluorescent dye molecule tagged with a short, complementary oligonucleotide sequence designed specifically for the associated PhenoCycler antibody.
- When the barcoded antibody and the reporter with the complementary oligo are incubated they undergo hybridization. This results in the generation of a fluorescent signal that can be effectively captured for each biomarker of interest.

PhenoCycler Antibodies For Human FFPE Tissue
PhenoCycler antibodies have been tested in multiple types of human FFPE tissue and are conjugated to a distinct PhenoCycler Barcode (BX000). Twenty-five (25) test scan be performed with each kit.
PhenoCycler Inventoried Antibodies for Human Fresh Frozen Tissue
PhenoCycler antibodies for human fresh frozen tissue have been tested using tonsil tissue. These PhenoCycler antibodies are conjugated to a distinct PhenoCycler barcode (BX000). Each kit contains one tube of PhenoCycler antibody and one tube of the corresponding dye-labeled reporter (RX000), sufficient volume for 25 PhenoCycler runs (or tests) at the standard staining conditions. Recommended dyes for human fresh frozen tissue are Atto 550, Cy5 or Alexa Fluor™ 647, and Alexa Fluor™ 488.

PhenoCycler antibodies for mouse fresh frozen tissue have been tested using spleen tissue and are conjugated to a distinct PhenoCycler barcode (BX000). Twenty-five (25) tests can be performed with each kit.
